Welcome to this introductory talk on general pharmacology. I'm Karel Allegaert. I'm a clinical pharmacologist located at KU Leuven, which is in Belgium, as well as in Erasmus Medical Center, which is in the Netherlands.

Why are we covering this topic? Simply because drugs are a crucial part of proven, effective interventions to improve patient health outcomes and healthcare efficiency, while individual responses to medicine may vary, related to medicine and patient-specific characteristics. That's all about PK and PD. From a health equity point of view and an outcome-relevant disease point of view, I can only provide you some examples, like, for instance, the recent COVID vaccinations, treatments for acute leukemia, or novel treatments for spinal muscular atrophy or cystic fibrosis, to illustrate the impact of effective pharmacological interventions.

What will we cover in these lectures? Well, we will cover, obviously, pharmacokinetics, and because of the volume, we have split this up into two parts. The first part is focusing on absorption, bioavailability, and distribution, while the second part has its focus on metabolism, excretion, and the therapeutic window. We will discuss routes of administration, drug formulation, pharmacokinetic and pharmacodynamic integration, and then a specific focus on pharmacogenetics and drug toxicity, including some mechanisms, interactions, and adverse drug reactions.
